Transformation Resources
Constructs:
A set of over 60 Gateway compatible transformation constructs have been developed at JIC. as part of the BRACT (Biotechnology Resources for Arable Crop Transformation) (www.bract.org) project. For barley and wheat, constructs are available for over-expression, RNAi and for producing 'clean-gene' transformants. Basic constructs are also available with different reporter genes including hygromycin and bar gene which facilitates easy re-transformation of previously transformed lines if needed. Promoters
A promoter catalogue is under development at RRes. So far, 22 constitutive, tissue-specific or inducible promoters have been validated in wheat transgenics using the GUS reporter system. Some of these are already incorporated into over-expression or RNAi transformation cassettes and are available for use. Others are available in cloning plasmids. This work is ongoing. Protein tags and targeting sequences
At RRes we have validated different immuno-reactive (epitope) tags for labelling 'transproteins' in monocot transgenics including His-, cMyc- and FLAG- tag sequences and the corresponding commercially available antibodies. In addition we have shown that GFP can be efficiently targeted to organelles such as the nucleus and various plastids in wheat transgenics. These constructs are available. (contact Huw D Jones).
