Transformation in Maize

IBERS

Contact: Sue Dalton

Current status

Maize hybrid genotype Hi-II can be transformed using particle bombardment with high efficiency and also with Agrobacterium using the bar gene for selection.

Costs and time scales

Depending on time of year 10-20 independently transformed T0 plants can usually be produced within six months. A further six months is needed to obtain T1 seed. Costs depend on the degree of collaboration.

Current work and future directions at IBERS

An experiment to compare two ubiquitin promoters showed that an enhanced rice ubiquitin promoter expressed more constitutively than a maize ubiquitin promoter and was not co-suppressed by the maize ubiquitin promoter used for the selection gene. Experiments are underway to improve Agrobacterium transformation efficiency and to evaluate new selection markers.
